Một thị trấn nhỏ ở miền Tây bắt đầu trở nên bất ổn khi một ông trùm xe lửa què chân muốn kiểm soát được vùng đất Sweetwater có đường sắt đi qua nên đã thuê các sát thủ xoá sổ gia đình Brett McBain. Cô vợ mới cưới của McBain là Jill trở thành người thừa hưởng toàn bộ gia sản. Cheyenne và một anh chàng Harmonica bí ẩn đã tự nhận nhiệm vụ bảo vệ Jill sau khi McBain bị giết. Nhưng sự việc càng trở nên phức tạp hơn khi anh chàng Harmonica bí ẩn này vô tình dính líu tới vụ tranh chấp đất giữa bà quả phụ Jill và chính nhà tư bản đường sắt tham lam, độc ác…
